I don't understand what you mean by "NPTH" layer.

You need to put V-cut lines on the Mechanical Layer:

Thanks for the suggestions, i have followed the same as above you  mentioned in the link, i have done the V-cut PCB and looks like as follows.

Now there is only One PCB section, remaining is empty, how to add the PCB to the remaining sections, Please guide.

Please read the Tutorial carefully and complete the order form to request panelization.

State that this is what you want in the Remarks box.

Then JLCPCB should do the panelization for you.

• Note however that your board may still be rejected because:

1. it is not possible to V-cut rounded corners;
2. It may not be possible for the V cutter tool to operate properly on anything other than a straight edge so your notched outer edges may not be acceptable for use with the V cutting tool.

I suggest that you email JLCPCB support directly to ask about this.
